Off the shelf packages tend to come under heavy analysis and the backdoors are not known for months after they have been exploited. PHP-Nuke is a prime example, I will only use that behind the firewall! mySQL has its problems, as do the majority of web servers used by domain hosting companies....
I would advocate building a site from scratch, using PHP and mySQL. Start out with something that has limited functionality but is useable ( e.g. tables for news, reviews, comments, users ).
Then add to it later.
